629 Validation of a wound fluid collection method for subsequent identification of prognostic biomarkers for healing

Several matrix metalloproteinases (MMPs) are hypothesized to play key roles in the wound healing process due, their ability break down various extracellular components. Expression levels of MMPs, or inhibitors, may be useful for predicting risk non-healing. However, methods used collect human fluid diverse with little evidence rigorous method validation literature. Therefore, we aimed develop a robust collection from dressings. We have validated this using both serum and samples. Serum was acquired commercially whilst dressings were collected ten patients undergoing treatment leg foot ulcers. Total protein concentrations relative abundances measured extracted control The developed gave normalised total recoveries (arbitrary units, a.u.) 0.64, 0.85, 0.60 0.56 a.u. when extracting alginate (Kaltostat), foam (Mepilex), surgical (Softpore) film (Tegaderm) respectively comparison controls (1.00 a.u.). Fluid patient had mean concentration 21.5 mg/ml consistent identification quantification several MMPs inhibitors via Western blot. therefore facilitated recovery minimal loss, specifically these data show variability between dressing materials. This method’s suitability further demonstrated successful extraction proteins involved healing. is suitable procedures development prognostic biomarker studies.
